The strategic alliance centers on diseases presently requiring injectable medications. Both companies aim to transform these injection-based treatments into oral formulations that patients can take by mouth.
Using its proprietary nGen platform powered by artificial intelligence, Orbis will identify macrocycles—ring-structured molecules capable of targeting biological pathways that conventional small-molecule compounds cannot effectively address.
According to Orbis CEO Morten Graugaard, the company’s synthetically designed macrocycles have demonstrated oral bioavailability levels reaching 18% in early-stage preclinical testing. This metric indicates the percentage of an ingested drug that successfully enters the circulatory system.
“Together with Novo, we have an opportunity to unlock a new generation of oral medicines for cardiometabolic diseases,” Graugaard said.
Financial Terms and Investment
The partnership encompasses initial payments, milestone-based compensation tied to development and commercialization achievements, plus tiered royalty arrangements on eventual product revenues. Specific allocation details of the $1.4 billion total remain undisclosed.
Novo Nordisk will additionally acquire a strategic ownership position in privately held Orbis. The financial magnitude of this equity investment has not been publicly revealed.
This partnership complements Novo’s current oral peptide delivery capabilities using SNAC technology. The pharmaceutical giant obtained this proprietary technology through its 2020 acquisition of American drug delivery specialist Emisphere Technologies.
A company representative from Novo indicated the organization remains committed to advancing innovation in oral drug delivery through combined internal development efforts and strategic external collaborations.
Industry Momentum for Oral Macrocycles
This partnership emerges amid expanding pharmaceutical industry interest in oral macrocyclic medications.
Johnson and Johnson (JNJ) secured U.S. regulatory clearance in March for Icotyde, an orally administered macrocyclic peptide treating plaque psoriasis. Merck (MRK) obtained approval three months later in July for Lipfendra, an oral macrocyclic peptide designed to reduce cholesterol levels.
Headquartered in Copenhagen, Orbis positions its nGen technology as purpose-built for accelerating macrocycle drug candidate identification through artificial intelligence capabilities.
The multi-target nature of this collaboration indicates both organizations will pursue opportunities across various conditions within the cardiometabolic therapeutic area, rather than concentrating efforts on any single disease indication or drug program.
